What is Nrf2, and how do you activate it?

Nrf2 is a protein, made from the NFE2L2 gene, that switches on your cells' own antioxidant defenses. Inside the cell another protein, KEAP1, holds it back; under oxidative stress Nrf2 breaks free, enters the nucleus and turns on the genes that make glutathione, HO-1 and NQO1.

How Nrf2 works

Nrf2 (nuclear factor erythroid 2-related factor 2) was isolated in 1994 (PMID 7937919) and is present in almost every cell of the body. Its job is a single decision: when to raise the cell's defenses. It works in three steps:

  1. At rest, KEAP1 holds it and destroys it. KEAP1 tags Nrf2 for the cell's recycling machinery, so Nrf2 lasts less than 20 minutes (PMID 15282312).
  2. Oxidative stress, or an activator, disables KEAP1. They react with KEAP1's sensor cysteines and the tagging stops, so Nrf2 builds up (PMID 9887101, PMID 14585973).
  3. Nrf2 enters the nucleus and switches genes on. Paired with a small Maf protein it binds the antioxidant response element (ARE) in front of its target genes (PMID 9240432).

Because the system works by releasing a brake rather than building something new, it can be switched on quickly (PMID 29717933).

What Nrf2 switches on

Nrf2 does not act on one gene: it regulates, directly or indirectly, more than 2,000 (PMID 36829925). The best-known are:

Mice without Nrf2 lose most of the induction of those phase II enzymes (PMID 9240432): that is how central it is.

Nrf2 is not an antioxidant. A vitamin C tablet brings one antioxidant molecule, used up the moment it neutralizes a free radical. Nrf2 neutralizes nothing: it is the switch that makes the cell produce its own defenses and keep producing them. That is why it is called the master regulator of the antioxidant response.

Why Nrf2 matters more with age

The Nrf2 response slows down over the years. In older animals, the Nrf2 in the nucleus fell by about half and the activity of the enzyme that makes glutathione fell 53% (Suh et al., 2004, PMID 14985508). The same signal produces fewer defenses, and that is why people switch Nrf2 on deliberately, with food or with a supplement.

Nrf2 activators: foods and compounds

Nrf2 is switched on by specific molecules, not by "eating healthy" in general. These are the best studied, and where you find them:

Food or a supplement?

Every item on that list switches Nrf2 on, and none of them sits on your plate at the amount used in the studies. Sulforaphane is not even in broccoli ready-made: it forms when you chew or cut it, and cooking lowers it. The broccoli-sprout drink of a 12-week trial with 291 adults carried 600 µmol of glucoraphanin and 40 µmol of sulforaphane a day (PMID 24913818): a concentrated extract, not a side of vegetables.

Nrf2 and epigenetics

Nrf2 does not change your DNA: it decides which defense genes are read, and that is gene expression, the ground of epigenetics. The link runs both ways: the Nrf2 gene can be silenced by methylation, and curcumin reversed that mark and brought Nrf2 back (PMID 21787756); butyrate, which inhibits the HDAC enzymes, raised Nrf2 and its protection depended on it (PMID 27799462).
